Control if the pattern of staining is appropriate for the antigen and applied antibody
A) Abnormal pattern of staining (granular) for CD30 in the cytoplasm of macrophages in case of classical Hodgkin lymphoma.
CD30. Granular cytoplasmic staining of phagocytized antigen in macrophages in the lymph node with classic Hodgkin lymphoma. Pattern of staining and cell type are not appropriate for CD30 staining.
CD30. Granular cytoplasmic staining of phagocytized antigen in macrophages in the lymph node with classic Hodgkin lymphoma. Pattern of staining and cell type are not appropriate for CD30 staining. Compare with normal staining pattern for CD30 in a single Hodgkin cell (to the right).
B) Abnormal pattern of staining for E-Cadherin indicating non-functional protein in a case diagnosed as lobular carcinoma of the breast.
E-Cadherin. Aberrant (cytoplasmic) pattern of staining in the lobular carcinoma of the breast. Notice normal membranous staining pattern in the epithelial cells of the small duct for comparison (upper left). 10x.
E-Cadherin. Aberrant (cytoplasmic, dot like) pattern of staining in the lobular carcinoma of the breast. 40x.
C) Compare the dot-like and diffuse cytoplasmic staining for Pan-Cytokeratin.
PANCK. Small cell carcinoma of the lung in the cell block from pleural effusion. Notice different patterns of staining “dot like” cytoplasmic in malignant cells and diffuse cytoplasmic in reactive mesothelial cells. 40x.
